In Ancient Egypt, Princess Ahmanet is the first in line to succeed her father Pharaoh Djoser. She has a love affair with Djoser's counsellor and polymath Imhotep. When Djoser discovers the affair, he has Imhotep executed. Ahmanet steals Imhotep's corpse and attempts to resurrect him but is captured by her Djoser's priests. As punishment, Ahmanet is whipped, has her tongue cut out, is mummified along with the men loyal to her, and buried alive with flesh eating scarabs. The curse turns Ahmanet and her followers into undead fields kept in a state of living death. Her sarcophagus is buried in Cairo and submerged in a pool of mercury so that her monstrous form will not escape.

In 1921, Joseph Morton leads an expedition to find the tomb of Ahmanet. Morton concludes the tomb is in fact a prison. He also finds the Amulet of Isis. Morton's assistant, Chris Vail, discovers the Book of the Dead and reads it aloud, accidentally awakening one of Ahmanet's followers, Kharis, who has now become a mummy. Kharis takes control over Vail and takes the book from him. While the sarcophagus of Ahmanet and her other followers are being moved to England, Vail attempts to open it. Greenway tries to stop him but Vail stabs him and attacks Morton, forcing him to kill Vail. The sarcophagus is returned to London while Kharis begins feeding on people in Egypt to regenerate his decomposed body.

Ten years later, Morton is now the curator of the London Museum where Ahmanet's sarcophagus is being kept. His son, Nick Morton, works in the museum with Jenny Halsey. Nick steals the Amulet of Isis and gives it to Jenny. Kharis, masquerading as a modern Egyptian Ardath Bey, arrives in London. Kharis travels to the London Museum and uses the Book of the Dead to awaken Ahmanet. Ahmanet begins feeding on people to regenerate her decomposed body.

While feeding on Joseph, Ahmanet encounters Nick and Jenny. She recognises Nick as the reincarnation of Imhotep and Jenny as a descendant of her younger sister who succeed Djoser after she after was mummified. She tries to convince Nick to join her but he refuses so she has her followers try to capture him and Jenny. The mummies are unable to go near Jenny while she has the Amulet of Isis, allowing them to escape.

Jenny later encounters Ahmanet, who has feed on enough people to be immune to the Amulet of Isis' effects. Ahmanet captures Jenny and sends her followers after Nick. Nick pursues Ahmanet back to her tomb in Egypt and is confronted by Kharis who reveals he also loved Ahmanet and was the one to betrayed her to her father's men to prevent her from resurrecting Imhotep, and now intends to kill Nick.

Nick is saved when Ahmanet kills Kharis for his betrayal. Ahmanet then has Nick mummified. Ahmanet tries to use the Scroll of Thoth to sacrifice Nick and resurrect Imhotep. Nick begins to remember his past life as Imhotep and various other people in history but is able to get free. Nick uses the Book of the Dead to resurrect Jenny and remove most of Ahmanet's powers, turning her back into a shrivelled mummy.

In a post credits scene, a weakened but alive Ahmanet is back in her sarcophagus, being lowered into a pool of mercury inside a Prodigium base. Prodigium's leader offers for Nick and Jenny to join his organisation. Nick continues to have memories of Imhotep's life and promises to Ahmanet that he will free her.

The second instalment, Bride of Frankenstein, would be released in 2019. Each of the individual monsters were stated to be the focus of stand alone instalments first, before any crossovers would occur. The 2014 film Dracula Untold, starring Luke Evans as the titular character, was originally considered to be the first film in the series; however since the film's release the connection to Monsters Cinematic Universe was downplayed and The Mummy was re-positioned as the first film in the series.
